I have never used that sort of box bt I do not think you should do that to a regional box
I like to send plain brown boxes by priority if I have to
I have many 6 and 7 inch cube boxes I use for 1st class but somtimes they go over 16 ounces
i prefer to use 5 inch cube boxes

As a rule the postal service does not allow you to change the intended use of special rate boxes. Even if they are being reused a second time.

If you don't wish to send it using reg rate postage, then use a plain box or a regular rate priority box.

I bet you can use that regional box with an express mail service if you want
I was always under the impression that priority mail boxes can be used for express mail since its sort of an upgrade
