When I use eBay's service for purchasing a shipping label, do I enter the dimensions and weight for my item itself; or do I use the dimensions of the box the item will go into?

when you use  "any label service" it is based on weight/dimensions for the item once prepared for shipping.

Including the box and all shipping materials.  Then weigh and measure.  I try to keep most things within 12x12x12 since shipping cost is the same as shipping 8x6x4, 8x4x4, etc.

It's the finished package that you have to weigh and measure.   It will be bigger than the item inside and it will add a bit to the weight.

Since the box is part of the shipment, you'll need to include the box dimensions and total weight. Good luck.
