My regular carrier is good about scanning my packages when she picks them up, but this week there's been a substitute. My media package wasn't scanned when it was picked up yesterday (Aug. 3) and the first scan shows Aug. 4 at 1:05 a.m. at my local facility. It should have been scanned on Aug. 3 to meet my handling time.
So, am I now going to get a late shipping defect? Or will I be okay IF it's delivered within eBay's stated time?

The same thing happens to me every so often, even tho I leave a BIG sign to ring my doorbell for pkg pick up!
So far I havn't rec'd a defect for those few pkgs bcuz they were delivered within the stated time frame.

My regular postal carrier SAID he scanned my pkg this past Monday, but it didn't show up until Tuesday after I told him I did not see the scan. The item was paid for on that previous Friday. I have a 1 day shipping/handling time.

I'm a bit paranoid as well and worry a bit more when it's media mail. I also have a 1 day shipping/handling time, but I'm going to change it to two. If yesterday's shipment does happen to generate a defect, I'll try appealing.

I've told my carrier the same thing and even threatened to call his supervisor telling her he doesn't deserve a vaca 😄

You should read the updates to the Ebay Seller Agreement. With the implementation of the guaranteed delivery program ebay has changed the rules for how the scans from carriers work and stop the clock on the seller handle time. You now have to take the package to your local post office, UPS, or FedEx location and have them scan the package there to stop the counter on the handle time. If you give the item to your local driver while on route when they scan the item it no longer stops the counter. Instead the counter keeps on counting till the package goes through a sort. They changed this because if you give the package to a driver on their route they might not get back before items ship out. This can cause a delay in the shipping of the item. For instance I get my mail delivered between 4 to 5 pm. The driver does not finish their route till like 6 or 7 pm but my post office closes at 4pm and packages received that day ship out at 4 pm to the local distribution center. So if I gave my package to my driver it would have a 1 day delay before it actually ships out to the buyer.

Having the driver scan your package may not always be bad as long as the package gets through a sort with in your listings specified handle time. However if the driver does not get it back before items ship out and there is a 1 day delay you will take a hit on your handle time. Then if you are taking part of the guaranteed shipping program if the item gets delivered with in the time frame quoted by ebay you are ok. However if the item is late and due to your handle time not being met if the buyer files a claim you will be charged for the shipping because ebay will refund the buyer the cost. For the time being the best thing to do is take the items to your local post office, usps or fedex location and have them scan the package.

I'm almost the last pick-up of the day too, but typically the packages seem to go to the distribution center after all the mail trucks are in. During busy times of the year, they'll often send drivers out just to pick up packages and get them in early.
Usually I don't run into problems unless it's a substitute carrier. They don't always scan the packages; they've even done a false scan on an incoming package. The driver this week missed delivering a package with the mail (came back later unbeknownst to me so the package was in the mailbox all night long). And he's also not delivered an important letter according to Informed Delivery. I'm really hoping my carrier will be back this week.
As far as eBay packages, I'll just increase my handling time a day and see how that works. It wouldn't be cost effective or a good use of my time to go stand in line at the post office.
