I am assuming the difference is one you use the flat rate box for, and one you do not.
I am shipping a shoebox w/ sneakers. What is the "large package" box? And will it fit my item?

I figured it out... I think. Large package goes in the "Large Mailing Box." And it is charged by weight.

Flat rate is usually used for expecially heavy items, USPS has priority shoe boxes

A "large package" is a box that exceeds 12" in length, width, or height. It is priced by size, weight, and destination. I think generally you provide your own box, but I believe the post office does have some available for free as well when shipping priority. Flat rate boxes allow you to ship anything that fits in the flat rate box (up to 70 pounds) to any US destination for a fixed price, insurance is extra of course.

Hi, Did you figure this out because I am wondering the same thing. I do not see a "large Mailing box" anywhere. Can it be any box?

Yes, you can use ANY box when shipping via Priority Mail. The maximum allowed length-plus-girth is 108 inches.
When setting up your listing's shipping calculator or the postage label purchase, just check off "Package/thick envelope" unless it's one of the special USPS flat rate or Regional rate boxes. Those special boxes will say "flat rate" or "Regional" right on them.
