When would be appropriate to file a not recieved claim

So an item that I expected to arrive today under ebays guaranteed date did not. Tracking is basically useless as it says label created usps awaiting item but they said they dropped it off. 

I filed a case with usps but that might not really do much if it is not actually going through the mail system 

If I file with ebay should I just say I want the item? When do I really file like today or immediately tomorrow?

I dont want to ding the seller but I also want to handle this right as Im out 60 ish right now so. 

Im sorry in advance for so many questions but im new to this and I dont know if just waiting on it like 1 or 2 more days is good or its best to file immediately

Whatever you do, the seller will never be out of the "60 ish bucks" as long as you give the money back to the seller should you receive the package after a seller refund.
