I purchased something yesterday on eBay, and it was paid for. Shortly after, I requested a cancellation. Turns out the seller is longer registered on eBay and the listing cannot be found. Is it still possible for the seller to cancel, or should I wait for after the est. shipping date to request a refund?

I have purchased/paid for items, then receive a message from eBay stating the seller is no longer registered or the item has been removed. I received the item with no issues. Never knew why the seller was no longer registered or the listing removed.
I would think you would be entitled to a refund from the seller regardless of their status and would be covered by eBay's guarantee. If that fails, you can dispute the payment on your credit card or PAYPAL account (maybe)?
