I recently bought a somewhat expensive electronic item (graphics card) that worked for two days and then malfunctioned. The seller specified "no returns," but would I be able to get a refund even if it was not technically DOA?

A seller's return policy covers remorse and is not involved if your item has a problem. You need to read ebay's money back guarantee to see if you are covered and follow the steps.

If the item failed, you should be able to file a succesful claim.
If something in your system caused the item to fail, that would be a different matter.

My wife's new car cost $56,000 and it came with a warranty to fix things that might go wrong. It's a warranty, not a return privilege.
Please show us the seller's listing where it details the terms and conditions of his product warranty and term of the warranty.
Yes Marge, I'm assuming the graphics card was used. Even if it was new in the box, why is the seller responsible for the "malfunction" when he didn't install it?
