How long are postage mailing labels good for? If I print a USPS postage label today must I mail the package today?

No, you don't have to mail it the same day, although it will get marked as shipped.

What ship date did you put on the label?
If you bought it today, you can ship it tomorrow without any problems.

The question isn't so much whether the USPS will accept it tomorrow instead (they will), but whether your handling time promises same-day shipping. The Handling Time clock starts when the package is paid for, and doesn't stop until the tracking number shows an in-network scan by the USPS, indicating that they now have possession of it.
If your local PO offers a Self-Serve Kiosk (SSK) in their front lobby and your package is not too large to fit in the drop chute, then you can stop in after hours, get a Provisional Acceptance scan at the SSK, and drop your box in the chute. eBay recognizes the Provisional Acceptance scan as proof of mailing. Your PO should follow that with a real Acceptance scan when the drop chute is emptied behind the scenes.
