My buyer did a return request. The item is media mail of 2 cassettes. He said it was the wrong cassettes. Can this go back through the Global Ship program?
If not, can I just go to Royal Mail and send him a label for postage? Ebay regular reps have not been able to give me an answer.

The GSP program is a one-way program and can't be used to return an item. You can't use the Royal Mail since the item will be returned from the US. You will have to send them the funds for return shipping.

No. If this is a Money Back Guarantee claim, then you are responsible to pay for tracked shipping from the UK to the USA.

If you are able to do that, sure. But, typically, an international seller gets a shipping quote from the buyer for tracked shipping, and then the seller sends money for the return to the buyer's PayPal account.
For low cost items, it is rarely cost-efficient to pay for return shipping on a Money Back Guarantee. You just refund the money ... if you believe the buyer is lying, then report them as abusing the Money Back Guarantee (multiple sellers reporting an abusive buyer will cause the buyer to lose return privileges or their account).
