I had one buyer buy one item on March 8th and then on March 10th buy another one of my items. Can I cancel their order or payment and send revised invoice combining the two items. Weight for both items is just bit over 1 once less than shipping each item separately. Will be cheaper for both of us as handling fee for one will not be added and I will pay less ebay fees. So many things have been happening in RL I can not think straight.

You don't need to cancel anything. Pack both items in one container, print off the shipping label for one of the two items as usual (with the updated package weight, etc., as needed), and then when that is done, go to your Sold Items page and mark the second item as Shipped. Finally, add your tracking number to the second item's sales record, and you're done. You can send a partial refund to the buyer for one of the two items to cover their overpayment of Shipping if desired.

Select both orders then print shipping label. The bulk tool will automatically offer to combine the orders if they're going to the same name and address and were purchased using the same eBay ID. Tracking will automatically be uploaded to both transactions.

Yes as the others have suggested, you can still combine the shipping.
If your buyer has already paid the full price for the shipping of both items, don't forget to refund your buyer the difference. Make sure you email them inside of Ebay and outline what the refund is for.
Don't combine the shipment and then charge your buyer the full amount without consideration of a refund.
