Always start the bidding at the least you will take.
If you want to sell it for $100, you have to start it at $100. Remember eBay fees will come out of that $100, too.

Good thing you asked and got a great answer from dryophelia.
If you started at $0.99 and it only received 1 bid, you'd have to honor the sale for a buck or use the out of stock cancellation option and get an account defect and probably some bad feedback from the buyer.
Don't use reserves either - expensive waste of money for something of that value.
Auction format is best when the item has enough demand to get bid up. If it's a fixed market value of $100, just list fixed price for $100 as tools suggested.

As others have said if you want at least $100 then start at $100.
Buyers can be a bit funny about round figures like $100 so maybe starting at $99 will get more attention.
