I don't ever do the "best offer" thing. I research my prices and decide what I want to get. I take into account previous sales, current listings, and shipping costs, and let that be that.
I figure if I want to do "best offer", I'll list it as an auction instead. Otherwise, I set my price and let it be. I can always come in and reduce it after a time of it not selling, if I want to do so.

I don't use the best offer listing. It's too easy and fast to find approximate values and what items have sold for. I don't want to waste my time with jibber jabber. I get too many emails as it is.
Anyone who makes and offer will expect an answer within minutes and leave feedback accordingly.

Typically your asking price is what you want for the item and if you choose a best offer option, it would be the price you would accept.
If you want to move a stagnant item than you may want to be more aggressive with a best offer.

I also do not use the "Best offer" feature. My experience was the people who would accept, wouldn't pay, so in my opinion best offers attract non payers just as much as auction style listings do. I would just post the price you feel the item is worth, no best offer and go from there. Like @toysaver stated, I don't have time for the random back and forth chit chat or price haggling.

I often use best offer when (1) I have some wiggle rooms in my price (margins) or (2) I have some uncertainty as to price due to lack of comps. But I make more sales by sending offers rather than by receiving offers.

Plus, if they haggle on the price before purchase imagine what they might do after.... They're already going into the transaction with some sort of dissatisfaction about pricing.

The nice thing about best offer is sometimes I'll set a price higher than I think I can get for an item with the anticipation that it'll lead to negotiating & accepting a more reasonable price, then sometimes someone will just come along and buy the item with no offer. I like when that happens. Also you can set a minimum offer price so you don't have to deal with low balls
