Im trying to get rid of some clothes in my closet, but I want to set a price that ensures that I make a profit.
Would it be:
Price + Free shipping or Auction at $.01 + Shipping

You wore it, you enjoyed it, you don't want it any more.
So your procurement cost is $0.00.
You will have listing costs.
Allow 18 cents a minute for your time photographing, measuring, describing and uploading. (Based on BC minimum wage.)
Then if you are listing fewer than 50 items a month, you have no Listing Fees.
But you will pay 10% of your selling and shipping cost as Final Value Fees and 30 cents +2.9% of your customer's payment to Paypal.
So forget the penny auctions.
BTW, Fixed Price listings make up 85% of eBay these days and most auctions include a Buy It Now price.
There's also the option of demanding Immediate Payment Required.
While most of the posters here mention that high end clothing is more likely to sell than cheap stuff from Old Navy or J.Crew, if you have Plus Sizes (18 and up) you will find more buyers even for ordinary brands. Check out prices for Eileen Fisher in Large and up, where used sells for very close to new prices.

Nice. And I buy most things used or pre-owned (to use a popular term). It has worked for me for many years. But I NEVER buy used mattresses. I just can't go there. And not related, but I also search out MADE In The USA.
