On all of my listings I have this one negative feedback that comes up first thing. It is hurting my business and I can't talk to a human being at Ebay. I am so frustrated.

Where are you seeing it show up as the first thing? On your feedback page, on a listing, where?
It appears you are getting close to having this one negative age off your percentage of FB. That is a good thing.
The one negative is unlikely to be causing you any problems with sales. It is unlikely it is hurting your business at all. Most buyers would just dismiss it. Not only is it old, it does not appear to be a repeat problem.

Your options will be on the left. If you use the link below you can only get to the Automated Assistant or Chat box type AGENT in the box and hit enter. You will then get more options. Not all options are available 24/7. It will depend on staffing available. So sometimes you can request a call back and sometimes you can't. Sometimes you have the Chat option available and other times it won't me. If it is important to you to use one of those options, just try back later.
If you use a cell phone or other mobile device, you may need to turn off your Spam filter so that Ebay can call you.
If you are a seller outside of the US or Canada, you will need to use the Chat Option.

The new way that "relative feedback" shows up when a buyer views an item is really a pain in the neck. Unfortunately, it seems that this new way that eBay has coded things, that negative may pop up forever when eBay thinks the feedback is "relative".

As a follow up to that
I clicked to view 10 estee lauder items, and some other items. (clothing, shoes, handbag)
I am not being presented with that neg.
I see a knit top, jaded gypsy skirt, and Dr Suess sweater positive feedbacks displayed

The behavior is really erratic. In addition, the mobile view offers a selectable choice popup between Newest First and "Most Relevant," but the desktop view does not. Selecting the alternate view (i.e. returning to "Newest First" instead of "Most Relevant") using the choice popup, if offered, doesn't stick anyway; you must select it all over again next time.
Simply refreshing the page can sometimes flip the ordering from one to the other, as can changing from All Received to Received as Buyer/Seller and back again. Also in "Most Relevant" mode, the subhead of "Detailed item information is not available for the following items because the feedback is over 90 days old" pops up semi-randomly when the first old over-90 feedback comment is encountered, even if that comment is followed by one or more recent ones for which item information is available.
If this was an optional revision or at least labeled as a Beta, fine, but simply rolling it out randomly and largely unannounced is not helping anyone. Not bothering to stick around and answer simple questions about it just adds insult to injury. Testing before release would have been preferable to hurrying it out the door in time for the Christmas rush.

Yeah, that's another complication. My Galaxy Z Flip3 5G phone can show what appears to be the desktop view via its browser, or the mobile view via the eBay app, and the behavior is rather different between one and the other. In particular, it looks like they remembered to add the Sort selection choice (Newest vs. Relevant) to the mobile app, but forgot to add it for the desktop view, even though the new ordering can be seen there as well.
One also wonders what the new code thinks is "Most Relevant" if the user has simply clicked on the seller's feedback count directly, rather than from within a specific listing.

I see some oddities even on my own stuff. Depending upon the item that I am looking at (mine) it may only show one feedback when scrolling down to where seller info is displayed.

That negative is on it's way out of your profile. If you look, it is under the 12 months column. It will only count against your FB score for one year. It will fall off soon.

One complaint about the new "Most Relevant" sort order is that it will cheerfully dig up Negative feedback from beyond one year ago, even though it's no longer mathematically included in the seller's FB percentage, and show it to a prospective buyer anyway.

I would just make a professional response to the FB, I do not think that it is hurting you as bad as you think, if I were vetting a seller and saw that they had only 1 neg I would likely not even read it and if I did it is one feedback and it would not stop me from buying
