I submitted to return something as did not fit my car. I received a shipping label from eBay. The seller asked me to close it the return & they would send me a label. I did that, but now they are not responding. Can I still use the original label that was supplied by eBay?

You could, but you may not get your refund since you allowed the seller to talk you into closing the return eBay won't step in to help you your refund because you closed the case.
You only other option is to file a chargeback with Paypal , if that is how you paid, or your credit card.
Bottom line never allow a seller to talk you into closing a return case.

What @ebooksdiva stated -- you should NOT have closed the case before shipping the item back to the seller, who would then refund you upon receiving the return.
Since the case is now closed, eBay cannot help you with the refund.

Thanks, guess I’ll chalk it up to a rookie mistake.

Yes, and even moreso the reason to use that label now that the seller is not responding.
The eBay return label doesn't expire -- you can check at the carrier website that it is still valid e.g. Pre-Shipment, USPS Awaiting Item -- and the seller will still be paying for that return label when it is used to return the item.
And now that the seller not responding after tricking you into closing the return at eBay, it is even more the reason to file
-- another return case the PayPal at this page if that's how you paid, or
-- a "returned but no credit" dispute with your card issuer at the card issuer website.
Just upload that tracking number on the label to show that it was returned @mee_5554 to get your refund.
