I got a bag from Japan, and I live in the US so DHL made me pay $250 import duty. Now I decide to return the item. Is there any way to get the import duty back? Will I be charged any customer duty again while returning? I really don't want to experience that again.😥

Rather vague in information. However, if you were assessed, the bag must have been over the valuation that requires the duty. Taking that into account:
You received the item via Importation, where legal import duty is owed before entry is allowed. That is a government tax & not eligible for refund. It was triggered by you as a buyer & not the seller.
As to if you will be charged duties for a return, in most cases, usually not. It is the responsibility of the receiver to resolve those costs, should they exist, on their end. Should they refuse to pay, item would be returned to you.
As to not going through that again - only way to avoid the charges are to buy domestic or within the US. Any good crossing an international border is subject to those duties & each origin country will have different duties due.

You do realize that if you decide to return this bag the seller would only have to refund you the original price for the bag and not the original shipping or return shipping unless you lie and state INAD which it is not according to your previous post. You are just upset that the customs fees are to high:

It was your responsibility to know what the Customs/duty would be on this $1180.00 bag. Just because you don't want to pay the Customs/Duty fee of $250 for a luxury item over $800.00 USD.

If I've found the correct item ... the Louis Vuitton tote bag ... that seller offers Free returns, so returning the item won't cost you anything. [You will have to wait until the seller sends you the money to cover the shipping cost, since they won't be able to provide a US shipping label.]
You do lose the $250 import fees you paid, however. There's no way to recover those.
