A bit confusing, but it sounds as if you are the buyer, and have opened a Return request... but now (for whatever reason), you want to cancel the Return and keep the item with no refund? If so, go into the Case notes and close the Return.
If you're the seller, you can't cancel the Return request. It's in your best interest to react to one, though.

Byer opened a return request before item left for him. he is now asking me how to cancel it. can you tell

It's important to make sure you're certain you want to cancel a request, as you won't be able to re-open a case once it's been closed.

turned down sellers offer of partial refund. Then through correspondence a deal was made on a replacement item. Want to cancel return request with eBay but can't because of my turning down partial refund request. How can I cancel the request now

It is not a good idea to cancel a return request. If the seller has asked you to close he could be attempting to con you and not send anything. Once the request is closed you can’t open another on the same transaction.
Keep it open until you receive either a refund or replacement. If you don’t receive the replacement as promised you can go back to the request and ask eBay to step in for a refund. If you receive your replacement the request will close on its own in about 30 days of no activity.
