I have an offer I'd like to accept. The shipping is $185 for a 90 pound item from San Diego to Florida. How do I send an invoice for the exact amount of my item plus shipping?

The shipping charges should have been indicated on the listing page, and when the buyer clicked on the Make Offer button, their offer was for price of item only, and shipping remains the same as the listing page. No need to send an invoice.

One more question please. My offer is from someone who just joined ebay today and has no feedback.
Should I be suspicious? I'm fairly new to selling here and have run into very few scammers.

Make sure they actually used the Ebay's Make Offer (not thru messages) and you accept through Ebay. If they ask you for your Paypal information & email address, it is a scam.
You only ship when Ebay sends you notification that the buyer has paid and OK to ship.

I also noticed that the shipping price listed is NOT the shipping price to Florida. Because the shipping is expensive, I listed examples of FedX Ground prices to different destinations and reminded potential buyers to calculate your shipping before purchase. I have the feeling I'm gonna have to cover some shipping here.

It is NOT up to the buyers to calculate shipping. it is up to the seller to have the proper shipping charges shown on the listing page (not in the description).
Whatever the buyer sees on the listing page regarding shipping charges is what they will pay. If you miscalculated or underestimated, then YOU will have to pay the difference out of your own pocket.
Since you have not accepted the buyer's offer, if you made a mistake with the shipping charges, you can decline their offer.
EDITED TO ADD: When you look at your listing, you are seeing the shipping charges for shipping to YOURSELF. The buyer should see shipping charges to THEIR location. For instance, I see $117.56 to MY location (same state as you)

I am located in 92802, and the shipping to me shows $106.85. I think going to Florida is going to cost a LOT more. I changed the zip code to 10010 and I believe it was the same cost as you had to New York, and most likely close to the cost to ship to Florida.
