When you enable Time Away, you get the choice to continue allowing sales (in which case eBay will automatically extend your handling time to whatever date you specify) or to pause sales (in which case no one can make purchases from you during your Time Away).
Note that there are some things that can still sell during Time Away, like auctions that end after your start date, pending offers, etc. It's best to make sure you give yourself some buffer for both the start and end dates to make sure you won't have any orders come through that you can't fulfill.
