What is a seller to do when you have said "no refund" in your ad still the buyer wants one?

As you have just found out , your no return policy means nothing and is trumped by ebays money back guarantee , perhaps you should go and read the guarantee now , this is a battle you cannot win .
You are not a newbie this is something you should know by now .

If the return reason is for an item not as described, your ebay return policy is trumped. If this is the case, tell the buyer to return for a full refund and send the buyer a return shipping label (which is required by ebay).

A "No Return" policy on eBay is basically useless, as buyers need only open up "Not As Described" cases against the seller, and then the seller not only must accept the return, but the seller is also stuck with the return shipping fees.
If a buyer has remorse having received an item, and wishes to return the item, the BUYER is stuck with the return shipping fees, and NOT the seller. Plus, the seller may ACTUALLY receive the item back for re-sale to somebody else. (Or maybe a box of rocks, or newspapers, or cat litter. . .or worse.)
Accept the return, and move on.
