Will eBay give me a ‘Seller Defect’ if I don’t upload a tracking number for a sold item?

Defect? Not really, just dinged for no tracking.
On another account I sell small paper items. I ship them in a regular envelope with a stamp. My tracking upload percentage is ZERO. My on time is 100%. The only way I could be marked as late would be if the buyer reported it as late. I've had no INRs filed ever on that account, and my seller status is above standard. (there are three seller statuses - below standard, above standard and top rated)
If there is no tracking uploaded or scanned, unless the buyer says otherwise by reporting it late or filing an INR, the transaction isn't even counted in your shipping metrics - only the tracking uploaded part.

Yes, under "Tracking uploaded and validated on time" it will add 1 to the quantity under "Not uploaded at all".

You sold an item. The final stage is for the Buyer to receive it. You can use a delivery service or local pickup. Your Sales Venue, your contract with Buyer, shows delivery options. If Buyer has paid for shipping, to fulfill your contract, you need to show delivery in a reasonable time. You need to have tracking loaded as part of contract, just for that reason
However, if you and your Buyer have agreed on local pickup, you don't need to have any tracking uploaded
Defects are given on basis of your actions to fulfill your contracts

Yes, it will but that's not the same as a Defect in the sense that eBay uses them to evaluate seller performance.

I completely agree, when I sold on eBay, I uploaded tracking numbers from the receipt at the post office - which is free! I personally felt that "my" buyers (when I was selling) deserved to have that info + I do not care for sellers who supply tracking - what if something goes wrong and you do not get what you paid the seller for?? When I buy from a seller who does not supply tracking I do not like that. I for one think eBay should step in and do something about this problem.
