I recently bought an item by a seller located in the US (I live in Athens, Greece), and the tracking number the seller sent me is ESUS129xxxxxx. I have never seen a tracking number like that. Does anyone have any idea what it is? Is it USPS?

The seller shipped your item using eBay International Standard Delivery @stylishbiologist. The ESUS number is a consolidated tracking number. Under the ESUS number is the USPS tracking number to the US hub. At that hub the package will get an international label with a new carrier and that new tracking number will be consolidated under the ESUS number.
The number will track on eBay but for more details than eBay provides and to get the local tracking number/carrier once the item leaves the US you can find more info on this site: https://parcelsapp.com/en

I've never seen that either, but have you tried putting it in the tracking space on the USPS website?

ESUS numbers can't be tracked on the USPS site.
ESUS numbers are currently assigned to 2 different types of shipments. The first is eISD as used on the OP's shipment and the other is ESE (eBay Standard Envelope) for trading cards, coins, postcards etc which is essentially a domestic metered mail service.
