On the day that I purchased a gift from eBay for my friend, he told me that he already had the item. I asked the seller for a cancellation (which I think is reasonable because it hadn't been shipped yet), and have not gotten a reply. If I don't get a reply from the seller, is there anything I can do to get a refund and stop them from shipping it?

If the seller has a return policy you can return according to their return policy

Yes, if your seller takes returns @katcal-36, as a courtesy, let it know that it will be coming back, so it can decide whether or to ship it.

How are you certain it did not ship before you asked to cancel? There are still sellers who take boxes to the Post Office with a hand written label and wait in line. The tracking number is on a receipt and has to be hand entered with no specific time requirement. It can ship days before the tracking number is added.

Seller is not required to cancel transaction all though i believe majority will if they can ( had not shipped item yet)
Also this is Sunday not all sellers check eBay on weekends s you may get a response Monday
but i feel i must tell you request to cancel will in many cases earn you to be added to a sellers buyers block list

there is no entitlement to a cancelation when making a purchase on eBay.

You deserve to know that cancellations cost sellers time and money and most sellers aren't too happy to see them! Sellers pay fees to sell here and we are charged fees again when items sell. The money is taken right away... GONE! Some of it we'll never get back, and to recoup a small portion of it, we sometimes have to spend time doing extra work to apply for the fees to be refunded. Sometimes, cancellations also work against our selling record with Ebay. They shouldn't, but they do, although they are no fault of our own! It's buyers who don't want to fulfill their side of the agreement who cause all this disruption and aggravation.
A lot of sellers would never tell you these things, but I feel buyers need to know it's no easy bed of roses to be a seller here under the best of circumstances, and worse when buyers waste our time and money. Also, sellers are under NO obligation to cancel a transaction as per Ebay rules. If you find a seller who will do that for you, it's a big courtesy, and you should be very grateful.
Keep in mind that seller will have to spend time dealing with your problem (as opposed to spending time with their family or productively working) and you will probably be blocked from bidding or buying from him/her in the future.
Maybe you should just await the arrival of the item and give it to someone else as a gift, or donate it to a worthy cause. You'll feel a lot better about yourself if you do.

And whatever you do... DO NOT file a return and say something is wrong with the item. Worst case, say you bought it by mistake or changed your mind. Saying something is wrong with the item will totally rub the seller the wrong way (myself included).

Users that try to avoid shipping it back at their expense get a rather amusing to me but rude to them awakening.

some sellers have hundreds of orders and if buyer place order on weekend and send message for cancellation then mostly sellers they print orders in bulk to make it sure it can be packed and shipped out, may be its a simple procedure in buyer eyes but its not that simple, some sellers print labels outside of ebay and ship it and then add tracking to buyer order. so in my opinion before placing order make sure what you are going to buy and make sure what is shipping policy of seller. second important thing once buyer place an order then paypal charge money from seller even then seller cancel it that money is not refundable to sellers. so always make sure if you are going to place an order that you have gone through all the details of listing.
as far as cancellation concern mostly seller does cancel the order if its not shipped but then there are chances you will be in block list of seller from future purchases.
