Do you have to use a Priority Express box if you print out an eBay label? Do you also require a signature with delivery because when I go to print the label, the box for signature required is selected automatically. Does this service guarantee one day delivery? A buyer just ask me last night if I should ship a small item overnight. They need it for Saturday.

Box, no.
Just like regular Priority, you can use your own box.
PME is not being guaranteed at this time, but I have had no problems with it. Just make sure that you get it to the post office and scanned before the cutoff time (my PO is 11 AM, yours may be different).
Some PME is 2-Day by default, depending on the delivery address.
Signature Confirmation is defaulted to "yes" on eBay, but the PO is waiving signatures due to "no contact" for Covid.
If you get it to the PO today, it should get there by Saturday.

Priority Mail Express is next-day delivery to most ZIP Codes, but not all of them. You can check the expected delivery time (including the cutoff time for you to get it to your Post Office) from your ZIP Code to the buyer's at usps.com.
If the buyer asked you last night, then chances are he also asked other sellers, who might have replied more promptly.
Given the difficulties with shipping lately, I would not take the risk of trying to go along with this buyer's request.

Signature confirmation is needed for items totaling $750 or more, to maintain seller protection. Otherwise it is at your discretion.
