The item would be in a USPS Large Priority flat rate box with all the priority markings.
Would Amazon accept this to ship overseas or do I have to use a plain box with label ?

If your item fits in a USPS Large Priority box you can certainly use that to ship to the Global Shipping Center in Erlanger, KY for ebay items.
Why did you mention Amazon in your post?

Dont forget to insert the packing slip before you check out. Ebay requires it.

I have never put a packing slip in a global shipment. They have all been shipped without issue. Do you mean the customs form?

You can ship either flat rate priority or calculated weight priority to the GSP place in Kentucky. Its just a domestic shipment for you either way. Dont use flat rate priority if calculated is cheaper and you have a plain non flat rate box that it will fit in.
If you are asking if you can use a used Amazon shipping box - yes although I would turn it inside out so it looks cleaner on the outside but thats just me - and a reused Amazon box cannot be used for flat rate priority shipping. Only regular priority shipping. If you are doing a flat rate priority shipment then you must use actual USPS flat rate boxes. Otherwise you can use any box that it will be safe in.
