We sold an item to a buyer that got delayed and the buyer was concerned about it so we contacted the post office and they said it was lost. This item was shipped via media mail so it could be insured so we replaced the item for the customer sending it priority mail to them and it was received by them yesterday. Now the original lost item must’ve been found and is now also going to be delivered to the same customer tomorrow. We requested that the customer return the second item to us but is there any obligation that they have to do that?

No. You can ask, but they cannot be required to oblique.
And here's the rhubarb: If they refuse delivery of the now-found package, you will have to pay Media Mail postage again, to receive it back. The Priority package would have returned with no additional postage, but it's already unpacked.

The customer isn't obligated to send the second item back. However, there is no harm in asking them to return the second item once the item reaches them. You never know, they might just ask to purchase the extra item from you.

If you want the original item returned, you need to file a package intercept.
I did it once and it worked...however, it does cost money so depending on how much value there is in that package, it may or may not be worth it.
