I am a relatively new seller with a store and have almost 200 listings of individual items (mostly women's clothes). Is it OK to create a listing for a "wholesale lot" of, say, 20 items and if it sells can I then cancel 20 of my individual listings (if they haven't been purchased) and use those to fulfill the order? Will I get a defect if I do this? Not sure if that's against the rules.
Thanks in advance for any advice.

Does it violate any policies? Not sure.
Its it an absolutely terrible idea that you will regret bitterly? Almost certainly.

You hold the right to end your listing at any time, but if you end the listing and people are watching or your items have current bids this will not look good for you, wait until all list end they re-list them as you see fit...good luk

It's OK to end a listing, you won't get a defect point. But if you list an item individually AND as part of a lot, you're taking a risk that the individual listing will sell, leaving you unable to ship the lot if it sells before you have a chance to remove or revise the Lot listing.

You would be listing the same one of a kind items together and separate to gain better exposure, not allowed.
Worst than that if one or more sold and the *lot* sold at about the same time you would not be able to fulfill one or more of the orders completely and would get an out of stock defect or a SNAD for not delivering the entire lot.
Which would do serious damage to your selling acct.
Only a few of those and would would be restricted.

It can be a troublesome issue. I know some sellers will list the same item on auction as well as a separate BIN. In this case, you run the possibility of the lot selling then one of your individual items selling at the same time (say overnight) and you will end up with an out of stock.
While the risk may not be that great, I don't think I'd take the chance.

Thank you everyone for your great advice. Thank you!

All your listings are fixed price. Yhere is no penalty for ending a fixed price listing.
However: what if one of the single items sells? Your wholesale lot is now incomplete. If someone buys it before you can cancel it, that’s when you’ll have a defect situation. And I’d someone buys the lot, you have to cancel 20 listings before someone buys one.
