I want to use my PO Box as my shipping from address. When I go to change it, on address line one do I just put my PO Box number and the city/town its in or do I need to specify a street address along with it?

FWIW- because some sellers will not ship to mailboxes, although those are the most secure addresses available, you may prefer to use
123 Anywhere Street, unit 456
Anytown NY 90120-0456 USA
instead of
PO Box 456
Anytown NY 90120 USA
I've googled a lot of customer addresses, and noticed that nearly half of the "apartment" addresses are actually in post offices or MailboxesR Us outlets.

You do not need to specify the street address of your post office.

I use a PO Box as my ship from address. I put just my business name (first line), PO Box 1234 (second line), City, State, ZIP +4 (third line).
For those who want to use a PO Box as a ship to address, then USPS offers the street addressing service as mentioned in the other replies. For example, if a post office is located on 123 Main Street and your post office box there is 789, your mail would be addressed 123 Main Street #789.
