To summarize, I sold a big pair of over-the-knee boots to someone for $30, and she paid $8.55 shipping for which ended up being a 3lb 8 oz package. I sent it priority mail (which ended up costing me an extra $2 more than she paid, but I was good with that because I underestimated weight) and shipped the next day, but it got stuck in transit somehow and wasn't delivered for 11 days. She opened an "item not received" on the 10th day and asked for a refund on what she paid for shipping only, saying "$10 shipping is WAY too much to spend for 20 day shipping" (it was only 10 days, and that was out of my control.) I told her to file a request with USPS and not me, since it was out of my hands at that point, to which she replied "USPS can't refund me the shipping cost, only you can. I understand that you shipped it on time but the package has not arrived on time. Please refund me the shipping cost, it's only fair. Thanks." I told her that I checked with USPS's refund page and entered the tracking number, but that it didn't qualify for a refund, and that once again she should contact USPS for anything further. She then said if I didn't refund her the shipping cost, that she would get her bank to dispute the entire charge. I did not respond to this because the tracking was updated that day saying it reached her town. It was delivered the next day, and she left positive feedback saying "Arrived in great condition. Shipping was way too expensive especially considering they were delayed 20 days (and the seller refused to help.) Otherwise beautiful shoes! Thank you". This feedback makes it look like I am uncooperative and did not ship for 20 days, however since it is positive I'm not sure it would qualify for removal (and I have never had to do that before.) Would you remove this, and is it possible to in this case? Thanks!

This particular feedback doesn't qualify as a false positive. The buyer was overall happy and stated so.
Let it go, nothing to see here.
Also DO NOT REPLY to the otherwise it will stick out like a sore thumb and make it even worse. Eventually it will be swallowed up in the rest of your positive feedback and no one will notice. It's just noticeable because it's at the top.

I would be happy you received a positive. If you really feel the need to respond, do so very carefully. Remember future potential customers will be reading it. I would respond with something like this:
Item was shipped the next day. Actual shipping charge was $10.55 via Priority Mail. I only charged you $8.55. Delay was due to the USPS. Thank you for your purchase!

Don't worry about it. First, it isn't really that bad and mostly aimed at the shipping company. Then, most people don't check feedback and when they do, they just look for negatives and neutrals.
If you continue selling the comment will be buried soon enough. Or, go buy a bunch of small priced items on eBay and wait for sellers to give you feedback to hasten that burial.

No one will ever read the words.

OP I would take it as a win that the package arrived before the INR could be closed in buyers favor, and also a win that they didn't file the chargeback.
The sheer audacity of this impatient buffoon to threaten you with doing a chargeback is appalling quite frankly. I wish people like this could be thrown into a huge meat grinder so the rest of us didn't have to risk EVER running into these Karens.
