Sorry I am fairly new to selling on ebay.
My auction recently ended and I sent an invoice to the highest bidder. A few hours later he messages me "I dont want to buy it" and now I'm unsure of what to do.
The 2nd highest bidder bid the same amount, is it possible to sell the item to them?

You can make a 2nd chance offer once the cancel goes through. Do not do so before the cancel is completed.

There are many times I would LOVE to get a 2nd chance offer because the high bidder backed out.

Yes, one certainly can do that. Its called an Unpaid item and can be set up automatically to occur as early as 2 days past closing OR manually do it through eBay's Resolution Cneter (that's how mine is set) ... BUT, in a lot of cases processing the UPI and then adding Buyer to your bocked list could result in losing a potential future Buyer. I always comply to a request to cancel then I can Relist immediately ... AND I always thank the Buyer for letting me know.

It does not any sense to open an unpaid item dispute. The buyer has communicated they do not want it. Not sure what more you intent to get out of doing what you suggested?
Cancel as requested and move forward past this event. No need to harbor it, or let it affect you moving forward and certainly no need to wait another 4 days for the dispute to close out.

Cancel as buyer requested.
You will not get a defect and you will get your fees back.
You can send a second chance offer to the next highest bidder, but those usually don't work out well.
Or you can then re-list.
