I see where Buy it Now requires immediate payment but does it alo mean the buyer can't make a claim against the sell?

Against the sale, you mean?
No. A buyer can still make a not as described claim or not received claim.

Immediate payment just means the listing will stay active until the buyer pays.
Claims can still be filed later.

I'm not sure what you are asking, but if you mean can a case still be filed against a seller if immediate payment is required then the answer is yes. When the payment is made wouldn't change anything about delivery or not as described cases being available.
