I just got back into selling on eBay and I am wanting it to become a good side hustle. However, from my past selling I had canceled 6 items and now since I don’t have many transactions I have a 31% defect rate. I will most likely become a below standard seller in the coming weeks and I am worried this will kill my chances of being able to get my shop off the ground. Is there anything I can do?Even though I have great reviews and as of recently I have been very reliable does eBay not care? Should I just open a new shop and start over?!?

Have you looked at your Seller Dashboard? That is what you need to do to determine if you are Below Standard or where your account is ranked.

Come back and tell us what you see. Now keep in mind if you are in the Below Standard level as a seller, you will be charged an additional 6% in penalty fees on top of your normal fees on Ebay.

I would just roll with this account since you only have the one negative feedback. Opening another account won't help you avoid the 6% penalty if you fall to below standard.
Just don't make any mistakes from here on out. One more defect on your account could be your last. And no, eBay does not care that you have been reliable lately. They could shut you down in a heartbeat. It is a numbers game with the metrics. No one from eBay is reviewing your account. A suspension would be by the eBay system, not a human.

6 canceled transactions made 6 buyers very unhappy with ebay and sellers, I can see one or 2 at most but 6? You need to manage your listings better, and it's not too late.

Not to mention a serious violation of the Ebay rules. To open another account in an effort to avoid the sanctions that are placed on your main account is a serious Ebay violation and could cause you to lose your selling privileges all together.

The best way to work out of this is MORE transactions completed successfully with your buyers. Stay away from posting your expensive stuff until you work you way out of the 6% penalties. Post low $$ value items, as many as you can. You want to dilute your numbers and to do that you need as many sales as you can get.
Plus Do NOT cancel anything unless you are cancelling and selecting the correct reason of the buyer requested it.
You still need to look at your Dashboard to actually find out what your issue really is and you haven't posted that you have done that. You came here for help. Some of us are willing to help you, but you MUST participate for that to happen.
