I am pretty sure you would need to start a new listing from the start. I know for sure if the active listing has any sales against it you can not use variations and will need to start from the beginning. It might be possible to add variations if there are no sales, but I highly doubt it.
Also not all categories of products have variations so you would need to check into that as well.
