It could be immediate; it could be never. (Sellers aren't required to respond, but many do.) I'd suggest that you don't buy anything until all your questions are answered.

I'd allow at least 12 hours. Sellers can be in different time zones and different countries. Also, sellers might work during (their time zone) day and might not be able to respond until after hours.
