Seller lists a “Best Offer” in addition to “Buy it now Price”. The seller didn’t respond before the 24 hr window expired. In fact, raised the “Buy it Now” price by $40 during my 24 hr bid. After the window expired, I sent a new bid for the item at the original Buy it Now price, but have since changed my mind in dealing with this seller waiting for the next window to expire again. I have attempted to contact the seller 3 times with No Response.
Is there any way to retract a Best Offer Bid during the 24 hr. window and be done with this seller ?

I think the seller isn't going to accept your offer anyway, but the answer to your question is on this eBay Help page: https://www.ebay.com/help/buying/buy-now/making-best-offer?id=4019 Scroll down to the "Canceling a Best Offer" section.

Your impatience is NOT a valid reason to retract an offer. Don't make the offer if you are not prepared to wait the full time YOU selected for the maximum expiration period (it also expires sooner if the listing ends or if it is an auction format listing and the auction gets any bid). The seller is under absolutely no duty to actively respond to your offer and letting it expire is a valid option.
Be aware that eBay adds Best Offer against the wishes of sellers and makes it difficult or impossible to opt out. Do not assume that the seller is eager for your offers.
