Item sold and paid by the buyer .the day I was going to ship it,I found some defect which I didn't know before.i contacted the buyer so many times but no reply from the buyer.what should I do

If the defect if significant or you just don't want to risk a return and you don't hear back from the buyer whether he'll accept it, maybe at a reduced price, you will have cancel the transaction.

It sounds like you are going to receive a defect regardless of what you do.  I would wait for the buyer to contact you.  S/he will most likely file an Item Not Received case, in which case you can state you sent X number of messages regarding the damaged item.  If you voluntarily refund the buyer, you will get your FVF credited, and a defect for the cancellation, but no defect for an unresolved transaction.
