A buyer requested in a note I ship a 400 item to an adress different than the one listed on eBay. I said I could not do that and promptly canceled the order. The order has now left me a bad feedback. Is there anything I can do about this?

Ouch. It would have been better to explain to the buyer that you would need to refund him, after which he could update his Ship-To: address in PayPal, then re-pay you with the corrected address. (He could also have just filed a forwarding order with his post office.) Unfortunately an outright cancellation like that did you leave you open to Negative feedback for it.
You might try a note to the buyer explaining this, and offer to take his payment again once he has fixed his address. Then ask for a feedback revision after that...

Just to clarify, if the order was placed with a shipping address in florida, and the buy request the order be shipped to new york by messsage, I must honor the new address?
From everything I read in the forum, sellers are recommended to cancel the order and ask the buyer to place the order again, which I did and offered.
I even offered a local pick up option since I live in the same final city. However the buy was in a foreign country, used a foreign phone number, and said his "cousin" could meet me to pick up the order. The whole thing sounded fishy so I canceled the order. Is there anything that can be done about this negative feedback?

I WOULD give eBay CS a call and ask them to remove the FB. I think there is a good chance they will do so. If you used problem with buyer's address (which it was) you will not get a defect.

I agree with a post above, call CS and explain the entire situation. They may remove it.
Since we have to deal with paypal and under them you are not completely covered by it, then one should not have to take the chance to accept it.

Oh, he's a scammer. I wouldn't worry about your negative. But check into feedback violations to see if the neg qualifies for feedback removal, and then ask eBay to have it removed.
Eh?
Asking for something not included in the listing... Change of address
Asking for something not included in the listing... Local pickup by his/her "cousin"

Thank you for providing additional -- and essential --details. The Florida address likely is a mail forwarder frequently used by buyers in South America. Nothing wrong with that. However, your buyer evidently noticed that you are in New York and therefore he/she could receive the order more quickly and with much less shipping expense. Asking for shipment to a New York address or to have a relative or friend pick it up sounds perfectly reasonable to me. He's actually doing you a favor -- cheaper shipping for you, faster delivery, less chance of a lost or damaged package. Personally, I'd go ahead and honor the request. If in-person delivery to an office is not convenient, then just mail the order to the New York address. Maybe even refund a bit of the postage if the difference is significant. During 20+ years on eBay, I've made such arrangements many times, as both seller and buyer. ~~C~~
The more compicated the shipping the more chance that item is going to get misdirected and you will be responsible.
And I certainly do not want anyone showing up at my home.
The address on the paypal account or you ask them to agree to a cancellation have them change it and and rebuy it.
