I'm new to selling on e-bay and a little confused. There's two ways to list your item. Quick listing tool or business listing tool. Apparently if you use the business listing tool you can imput sales tax. I thought sales tax is automatically charged no matter what tool you use because it is a marketplace site, except in Missouri where they need to submit their own sales tax. I want to make sure sales tax is charged for whatever state I sell into. Does it really matter what tool I use? Or must I select the business listing tool and choose the sales tax for my state? Maybe this is just an outdated glitch with the business listing tool and sales tax is is charged no matter what?

You don't need to do anything. Ebay automatically collects and remits tax for buyers in states that require it.

You don't need to worry about sales tax. Ebay handles it for you.

I live in Missouri. Most of my sales this year have been to other states with a few into states that eBay does not collect and remit sales tax for. I want to make sure I understand... Since MO is not on the list of states that eBay collects for do I need to 1) figure out how to collect sales tax for all the transactions I had sell since the state I'm in isn't on the list or 2} collect and remit sales tax myself only for the states that aren't on the list aka only a few transactions? and if #2, do I need to apply for a sales tax remit license?

eBay will collect tax for all states that require it. Tax is NOT collected if the state does NOT require it. [Missouri and a few other states do NOT require tax be collected … yet.]
You do NOT have to collect tax for anything you sell on eBay. You do NOT need a tax license.

You don't collect sales tax therefore no need to report something you aren't responsible to collect and report.
