I am for the first time choosing to print my shipping labels to save on the S+H costs. I mostly sell clothing and reuse poly bags I receive from retailers when I order clothing for myself to ship items I sell. My question is, the polybags are all various sizes and once the clothing article is put in them it obviously changes the shape of the bag so am I okay with just inputting the package weight and not the dimensions before I print the label?

If the form requires you to enter dimensions, just enter a rough estimate or carry over the dimensions of the previous package. They don't really matter unless you're shipping via Priority Mail and the package volume is over 1 cubic foot (i.e., 1728 cubic inches).

I have no way of checking the previous dimensions. I did a very rough measurement of the length, width and height mainly counting from the shape of the folded clothing inside the poly bag and not the actual corners of the bag. I am just worried the package might get rejected and I will have to pay for shipping all over again.
Do I need to measure thegirth as well? I selected the large package option, it wieighs 3 lb 15 oz

As long as you have the correct weight, you should be fine.

it's actually 3 lb and 11 1/8 oz but I rounded it to 15 oz shold I just round it to 12 oz?

I meant "previous dimensions" as a convenience, i.e., using whatever carried over onto the listing form from the last item you listed.
As noted above, a rough estimate is just fine. The length and width of your envelope are based on the distance between the "corners," not on whatever is inside it. Your package is not going to be rejected as long as its actual dimensions are within the allowed range for the service you purchase. For Priority Mail that's one cubic foot. No, you do not need to measure the girth; the shipping calculator will do that based on the dimensions you enter. There is no reason for you to choose "large" package.

You are totally overthinking this. Priority Mail is priced in full pounds. The postage cost is the same whether you enter 3 pounds 11 ounces, 3 pounds 12 ounces, or 3 pounds 15 ounces. It absolutely completely does not matter which one you enter.

I've used poly bags and Padded Flat Rate Envelopes and 6x9 brown bubble mailers and each and every time I print my labels on eBay (by clicking the print label button on my sold page) the label prints exactly the same size. A nice 4 x 6 1/2" label. Both the label and the Online Label Record (that's the one I cut from the label sheet and keep for my records) come on one 8 1/2 X 11 piece of paper. All I have to do is insert the ounces/weight and click Print.

@grouch_douglas, Have you ever ordered the free Padded Flat Rate Envelopes from USPS? You can ship very heavy items in those and pay one flat rate.

@grouch_douglas - I shipped one out the other day and it was $6.90 my cost, including my seller's discount. If you want to order them, sign up with USPS and then find their Priority Mail Padded Flat Rate Envelopes and have them shipped to your home free. They come in bundles of, I think, 15 per package. If you have difficulty finding these free envelopes there, just type this into their search box: EP14PE_X
They measure 9.5 x 12.5

9.5 x 12.5 is not big enough. The clothing I sell is typically thick hoodies. Even folding them as compact as possible it still would not fit
