My buyer did respond to confirm the address , but said it is really a PO address. I know to send it to the address given but: does the PO put it in the PO box anyway and does he have time to change the address with Ebay befor I send it?

Do your homework.
1. Review the buyer's complete profile. Their location and all 3 feedback categories.
2. Google map the address to get an idea. You can map and satellite which is probably more current.
3. If international, I would cancel.
4. IF you do ship within your country, pay extra for signature required at delivery.
5. You could also send another e-mail to buyer and ask why they made that comment and then purchased. This may help with your decision, and it creates a communication thread that eBay can see. If they don't respond, you probably have your answer.
6. Could it be a UPS store that provides mailbox service, and the buyer is not providing that box number? I had that issue before with a buyer.

You ship to the address you received with the payment. Nowhere else. If he wants to change the address to have it sent someplace other than whatever his primary eBay address is, he needs to do that before sending his payment, and then select that address when he completes the transaction.
Bottom line: you do not take messaged requests to send anywhere else; you stick firmly to the address you receive with the payment itself, which will come to you through eBay, as shown on your Sold items page.
Incidentally, one common scam is to use a throwaway eBay account to send messages to sellers of any recently-sold item, asking to have the shipment redirected to the scammer's address, and hoping that the seller does not notice that the redirection request did not come from the ID of the real buyer. Make sure you are exchanging messages with your buyer, not a different ID.

Post Office clerks are usually literate.
It's practically a job requirement.
And most are smart enough to "translate" a disguised PO box address.
So if what you are seeing is :
123 Main St, Unit 456.
The clerk will read Station A, 123 Main St., PO Box 456
And put it in your customer's box.

It doesn't matter if they change their address, it doesn't change the address associated with the order and that is the only address that matters.
