I am being charged with Insertion fee for every new Listing. My listing total is below 250 and I also have Basic store subscription. Can anybody please let me know, why Ebay is deducting 0.3$ insertion fee on each new listing?

Have you ended and relisted items this month? Every time you list something, regardless of what ends up happening with it (ended, sold, removed, etc), it counts as one of your free insertions.
If you go to your Seller Hub Overview page and look for the Promotional offers tile there, it should show you how many free insertions you have remaining.

You may be adding new items to your store that are not included in your allowable inventory -- not everything can be listed with zero insertion fees.
Check the information which eBay had provided you, at the time that you had enrolled your Basic store -- you will probably find a list of limitations for which you will have to pay a 30 cents insertion fee -- and maybe 40 cents.

Are you shipping from Pakistan or dropshipping from Amazon, which is not allowed.

On eBay.com, the USA eBay site, the insertion fee is 35 cents ($0.35).
That applies, for non-store sellers, only after the seller has more than 250 listings.
Are you listing on eBay.com, or on some other eBay site centered in another country?

You are allowed 250 free listings per calendar month. If something sells and you list another item, that uses up one of your free listings. Every listing you post goes against the 250.
Watch your Seller Hub. There is a tile that will tell you how many listings you have left on your free listings in the Promotional Offer Tile.
