The buyer for my item hasn’t paid in 48 hours, they say the can’t pay on eBay and need to do PayPal or bank transfer.
I put collection only and they said they were coming, now they are talking about a courier.
I feel uneasy as it’s not a cheap item. I just wanted someone local to get it and come collect and I’d know they were happy etc.
can I cancel or will I be charged?

If you give the buyer your contact info you will receive a very official looking paypal email stating it's been paid. I would recommend cancelling the order, use the reason "problem with buyer's address" which we are allowed to do in cases where the buyer is asking for something "extra" (fake payment off eBay in this case).
If you do choose to relist, list it as a Buy it Now/ Good til Cancelled and click "Immediate Payment Required". However you may want to sell local for cash only.

If you are in the USA you can now open an Unpaid Item Dispute in the Resolution Centre at the bottom of this page.
If you are a non-US seller, you must wait 96 hours (four days) to open the Dispute.

Even if you are signed up for Managed Payments, PP still feeds Managed Payments, so the buyer is either lying or none too bright.

BTW - I understand that Managed Payments also allows ApplePay and GooglePay. I have no idea what either of those are, and I don't want to.

Bank transfers (wire transfers ) are common and accepted in Europe.
If you do not ship to Europe (which is likely where this guy is) you can cancel as Problem With Address.
You can refuse a bank/wire transfer as they are very very expensive in North America (your bank would charge you $25 or more to process it) and the seller chooses what payment she will accept, within eBay's parameters.
End communication.
Do not Ship.
Open the Unpaid Item Dispute.
Close it after 96 hours (US or non-US).
Do not believe any confirmation of payment that is not in your Paypal or Managed Payments account.
You can buy a shipping label from a Held PP payment. Held payments are normal with new sellers. Don't panic.
Cancellations are faster, but the "buyer" can leave feedback and there is no damage to her account.
With a Dispute the deadbeat gets a Strike.
You can set up an automatic Block on deadbeats with Strikes through your Seller Preferences/Buyer Requirements.

The buyer is trying to STEAL from you. Whatever they were to send you would be a FAKE payment notice.
You tell them that since they have purchased the item they need to pay via the payment terms during check out on Ebay that is the ONLY way they can pay.
Don't fall for this. It WILL end badly for you.
