I've encountered my first return that I have to pay return shipping on.
Do I use the same tracking number and request a return label
Do I create a label from scratch using my USPS Account (never tried)
What's the best way to send it to my customer?

You can select the eBay generated return label in the Return case that the buyer opened - those labels do not include insurance, if that's something you need you can purchase a return label from another source (Pirateship.com, Paypal)
From eBay:
If the package meets the size requirements set by the return carrier, you can purchase a return shipping label through the return request and send it to your buyer. They can then print the label and use it to send the item back to you.

Or you can supply your own return shipping label if you prefer so it has insurance
