I am sending an item total weight 6 ounces.
Out of habit, I created a label for 4 ounces, the usual weight for items of this type.
Since the post office is able to charge sellers for insufficient postage, do I necessarily have to void the 4oz label and buy a new 6oz label?

If you have not already shipped it then YES you SHOULD. It might slip through BUT if they catch it it is probably going to cost more in back charges than what it would cost you to do it right in the first place. Although I admit I do not know for certain, the post office uses posted rates whereas you buy it here at a discounted rate. The difference in the posted rates (in dollars) may be larger than the difference in the discount rate.

Sort of 'off-topic', but remember there are price breaks at 4, 8, 12 and 15.9 oz. You could put 8 oz. on the 6 oz. package for the same price. That's what I do with mine, anyway...
