Yes it could effect your shipping times if is beyond what you have on your listing.
Did they request it after they purchased it and agreed to the terms of the listing?
If not, you could always change your shipping times on the listing.

It was requested after payment as they will be on vacation

Nah, this type of thing needs to be communicated BEFORE the sale which would have allowed you to alter the handling time. All the buyer has to do is file an INR once the estimated delivery date has passed and yes, it will affect your rating.
If it were me, I'd bite the bullet and cancel and refund. Let the buyer know that you will relist with the proper handling time and they can repurchase the item if they wish.

If you ship quickly on your orders, typically 1 order that is shipped longer will not hurt your shipping ratings. If they are communicating with you via Ebay messages and seem legit, there is nothing wrong trying to work it out for them. If they put the message on the invoice notes and will not respond back via Ebay messages, you do have a case to cancel the order. But before you do that, i would reach out a few times over 24-48 hours (especially if they are on vacation, may not have access to computer) to let them know it will be cancelled if they can not agree (Via Ebay Messages) to what the listing states.
If you are shipping USPS, you could also ask them to go online and put a Vacation hold on there mail. This way you can send it and USPS will hold there mail at the post office until they get back.
