You must know what the shipping weight, and size is.
You must know where it is going.
You must know what carrier you are going to use to ship.
Check the carrier site for cost.
At that point it is a piece of cake.

Before you sell the item you can put the box size and weight in the listing and choose calculated shipping. Then the rest is taken care of for you. When you make a sale it will automatically calculate the price from your zip code to the buyers.
After the sale you can go to the USPS website and calculate the cost with the 2 zip codes, package dimensions, weight and type of service.
If they already made a purchase and you print your own labels fill out the form without buying the postage. The actual amount will be in the bottom right on the screen.

You can use the eBay shipping calculator, linked below. Most services are based on package weight and distance, so it's impossible to know the item's shipping cost if you don't already know the buyer's location.
