The 1st thing you should do is check the help pages and determine if the item that you are selling in the category that that item belongs is allowed to list using multi-variation style listings.

I sell books. The book category on ebay does not allow multi-variation listings at this time. 

I hope that that changes here in the near future.

I don't use Variations, but I do know that you have to start by using a brand new listing.  You cannot take a current/past listing and revise it.

What you can do to save time and effort is to revise the existing listing and just copy the HTML code to a brand-new listing that allows you to setup variations.

You can not change an active listing into a variation listing.  You need to create a new listings using the advanced listing form.  The variation option at near the top of the form ( 2nd or 3rd section from the top).
