Purchased a pair of recessed ceiling lights, one is defective (frayed wire).
Opened a return, but realizing that the return was for a refund of whole amount, canceled.
I'm just looking to get a replacement for defective item (partial return).
Asked the seller, they suggested contacting eBay since they are new enough that they don't know how to go through that.
Hunted all over their help pages, can't seem to find a link for contacting a human person to explain this (I'm sure by design).
Q1: Does eBay have a mechanism for allowing a replacement of a purchase?
Q2: Does eBay have a mechanism for allowing a replacement of part of a purchase?
Q3: Since my return is closed, am I SOL?

you have 1 chance for the return. You closed it and now eBay can not help.
To get a refund you would have to returned the whole order.
There is no "replacement" only return / refund.
Since you are talking to the seller. The seller should be able to go into the sale and refund you for the 1 item.

If you paid by credit card a you always should. Contact your card company to file a case.
If you had not closed the case, eBay does allow replacements. However, it's always best to just get a refund as you might net be happy with the replacement either.

Thank you everyone for your input.
After starting the process, once I saw that eBay was trying to process a refund for the whole amount with the QR code to take to USPS, I tried to back out of it and hitting the Cancel button did (with one final confirmation) close out the return, and with it any leeway from my end. This was one of those "I gotta pick something, and no matter what I choose, it's going to be the wrong selection" type of things.

Once you had sent the return request, I'm fairly sure that the seller would have had the option to offer you a partial refund or to send a label so that you could return the item.
