Generally you want to offer your buyers the most cost effective shipping possible. This is usually usps ground advantage, unless the item is large or heavy - then ups or fedex might be better choices. But you can also choose what works best for you - I am not willing to drive an extra 15 minutes to offer shipping that is $1 lower.
When you list your item and click add service under shipping, you should see the range of prices for a number of services, so you can make your choice. You can offer more than 1 choice, but pick the lowest cost first so that is what shows in search. I often offer both ground advantage and priority mail.
When you sell an item, you want to send by the same method you put in your listing that the buyer selected. Never send by a slower service than your buyer paid for.
