What happens when a seller doesn't accept offers from you?
I went to purchase an item from a seller and got a response this seller is not accepting offers from you...

It sounds like the seller has added you to his blocked buyer list.
This could be for all sorts of reasons (for example, you've made one or more lowball offers, you've made unsolicited offers meaning you submitted an offer on a non-Best Offer listing through messaging, the seller was uncomfortable with a previous transaction or communication, etc.).
The seller will almost certainly not explain the reason to you, so you'll have to move on and find a different seller.
